Abstract
Objective To explore the protective effect of Qingyangshenylycosides (QYS) on hepatic injury induced by carbon tetrachloride (CCI4) in mice and its mechanism. Methods Sixty healthy adult mice were equally randomized to six groups. The mice in control group and model group were given same volume of normal saline intraperitoneally, once a day; those in dimethyl diphenyl bicarboxylate (DDB) group were given intragastric administration of DDB, 150 mg/(kg · d); those in low-dose-, medium-dose- and high-dose-QYS group were given intraperitoneal injection of 20, 40, and 80 mg/(kg · d), respectively, lasting for seven days. One hour after the last medication, the mice in all groups, except the control group, were intraperitoneal injection of 1 mL/L CCI4 peanut oil solution 10 mL/kg to create a hepatic-injury model in mice. Sixteen hours later, the eye balls of the mice were removed, blood taken, the mice were then killed and liver collected. Serum alanine aminotrasferase (ALT), aspar-tate aminotrasferase (AST), interleukin-6 (IL-6), tumor necrosis factor alpha (TNF-α), the levels of superoxide dismutase (SOD), malondialdehyde (MDA) in liver tissue were measured. A pathology examination for liver was conducted. Results As compared with the model group, the levels of serum ALT, AST, IL-6, TNF-α and levels of MDA in hepatic tissue in each QYS group were notably reduced, and the activity of SOD increased, the differences being significant (F=2. 766 - 23. 952;q=2. 161 -9. 305; P<0. 05,0, 01) , the damage of liver lightened as well. Conclusion QYS has a protective effect on acute damage of liver in mice induced by CC14, the mechanism might be related to anti-oxidant and decrease of releasing inflammatory cytokines.关键词
