Abstract
To observe the change of large calcium activated potassium channels (BKca) in the rate with detrusor instability (DI) and discuss the role of BKca in DI. Methods Model of DI of female rate was prepared by ligating the proximal urethra in the perimeum. The single bladder smooth muscle cell was acquired by digesting the bladder smooth muscle with enzyme. Whole-cell BKca currents were recording with perforated whole-eel congfiguration. Results There were plenty of BKca currents detected in bladder smooth muscle cells and these electrophysiological properties ere consistent with BKca in other tissues, including outside rectification and being blocked by IbTX, a special blocker of BKca The amplitude of BKca in DI group was decreased in several membrane voltages. The current density at Vm= +60 mV was decreased to (20±4.63) pA/pF (n = 18) , compared with the control group (32.42 ±7.97) pA/pF (n=21) , P<0. 01. Conclusion BKca currents were decreased in rat DI, which may be involved in the DI and BKca may beome one therapeutical target of DI.关键词
