泄浊解毒方对溃疡性结肠炎大鼠TLR2及IL-8 IL-1β影响的研究
The Effects of Xiezhuo Jiedu Decoction on the Expression of TLR2, IL-8and IL-1β in Ulcerative Colitis Rats

Abstract
Objective According to the pathogenesis of ulcerative colitis (UC), we composed Xiezhuo Jiedu Decoction, the effect of which was purging turbidity, ascending lucidity, detoxifying and removing blood stasis. We carried out the animal experiment to explore the effect of Xiezhuo Jiedu decoction on TLR2, IL-8, IL-lβ in ulcerative colitis model rats, and elucidated the possible mechanism of this decoction on ulcerative colitis. Methods: 2, 4, 6-trinitrobenzene-sulfonic acid (TNBS) / etha-nol solution was administrated by enema to make ulcerative colitis rats models. Five groups were divided in the experiments: normal control group, modeling control group, Xiezhuo Jiedu Decoction high-dose group, Xiezhuo Jiedu Decoction small - dose group, Salazosulfapyridine group. The expression of TLR2mRNA was detected by RT-PCR; the expression of TLR2protein was dectected by immunohistochemical staining; the contents of IL-8and IL-l|3were detected by radioimmunoassy. Results: Xiezhuo Jiedu Decoction could significantly reduce the expression of TLR2, IL-8and IL-lβ. Conclusion; Xiezhuo Jiedu Decoction has good therapeutic on decreasing the expression of TLR2mRNA gene and TLR2protein, the content of IL-8and IL-lβare also decreased. Its mechanism may be related to reduce the expression of TLR2, IL-8 and IL-lβ.关键词
