毒胡萝卜素对ReNcell CX 人神经干细胞存活、增殖和凋亡的影响
Influence of thapsigargin on the survival, proliferation and apoptosis of human neural stem cell
摘要
Abstract
Objective To investigate the influences of thapsigargin on the survival, proliferation and apoptosis of human neural stem cells. Methods The human neural stem cells ReNcell CX were randomly divided into two groups, the experimental group was treated with thapsigargin of different concentrations( 10, 20, 50 and 100 nM respectively); the control group was given no drugs. The LDH release was measured via Spectrophotometer method, the cell proliferation and Caspase-3 level were obtained by immunofluorescent stain. Results The LDH and Caspase-3 level in the experimental group were significantly higher than those in the control group, thapsigargin inhibited the proliferation and induced the Caspase-3 release of the ReNcell CX stem cells in dose-dependent manner(P < 0. 05, 0.01). Conclusions Thapsigargin can inhibit the survival and proliferation of ReNcell CX human neural stem cells, which may be caused by promotion of ReNcell CX human neural stem cells apoptosis via activation of Caspase-3.关键词
