Effect of Qutan Chushi preparation on the expression of TNF-α, IL-1, IL-6 mRNA of chondrocytes in osteoarthritis patients in vitro

Abstract
Objective To investigate the effects of Qutan Chushi preparation in the treatment of osteoarthritis (OA) and its mechanism. Methods Chondrocytes from the resected knee joint cartilage of patients with OA were isolated and cultured, cells of the third generation were divided into seven groups: A group was incubated with DEME as blank control; B, D and F group was incubated with 5% , 10% and 20% Qutan Chushi preparation serum respectively; C, E and G group was incubated with 5% , 10% and 20% NS serum respectively. After 4, 8, 24, 48 and 72 h, the expression of TNF-α, IL-1, IL-6 mRNA in chondrocytes of every group were measured by real-time polymerase chain reaction (RT-PCR). Results Compared with A group, the expression of IL-1, IL-6, TNF-α mRNA in B, C, D group decreased, especially which in D group, the down-regulation of the IL-1, IL-6, TNF-α mRNA reached the peak respectively at 8, 24, 48 hs after the treatment of medicated serum. Conclusions Qutan Chushi preparation of 10% medicated serum can be used for the treatment of OA, the mechanism may due to reduce inflammation and delay the degeneration of articular cartilage cells through influencing the expression of IL-1, IL-6, TNF-a and some relevant genes.关键词
