Effect of cofactor regulation on succinate production in Escherichia coli during dual-phase fermentation

Abstract
Two strains, including E. coli NZN111 and its recombinant strain E. coli NZNlll/pTrc99a-pncB were investigated as the production of succinate. During dual-phase fermentation, E. coli NZN111 generated succinate as the main products, meanwhile, pyruvate was accumulated. With the overex-pressed the nicotinic acid phosphoribosyl transferase, the recombinant strain lowered the concerntration of pyruvate, eliminated the generation of acetate and increased the production of succinate under dual-phase fermentation. The yield of succinate and the glucose consumption rate was increased by 139% , and 20% , respectively.关键词
