Abstract
Objective To explore the regulatory effects of dexamethasone on bone marrow macrophage cultured by M-CSF in vitro. Methods M-CSF-induced bone marrow macrophage were cultured with the different concentrations of dexamethason.and the morphology and the cell proliferation were observed. Immunofluorescence method was performed to examine the phagocytic function,and RT-PCR technique was used to measure the mRNA levels of inflammatory factors. Results Proliferation of the cultured cells was inhibited by dexamethasone in a dose-dependent manner. The phagocytic functions of M-CSF-induced bone marrow macrophage were effectively inhibited by dexamethasone. The cultured cells were oval like,and their pseudopod became wide and short. Dexamethasone inhibited the related pro-inflammatory factors in the cultured cells. Conclusion The results suggest that dexamethasone could inhibit the activity and function of M-CSF-induced bone marrow macrophage in vitro,which provide an experimental basis in the treatment of hemophago-cytic lymphohistioeytosis.关键词
