摘要

Abstract

Objective:To investigate the relationship between thymic stromal lymphopoietin(TSLP) and unexplained recurrent spontaneous abortion(URSA) . Methods:The mouse models of spontaneous abortion (CBA/Jx DBA/2) and normal pregnancy(CBA/J xBalb/c)were established. The expression of TSLP in chorion and decidual tissues was screened by RT-PCR .Western Blot. ELISA was applied to detect expression of cytokines TSLP .cytokines IL-4 and IFN-y from peripheral blood. The expression of FoxP3 was detected by FACs. Results: Real-time PCR analysis indicated that the expression of TSLP in chorion spontaneous abortion model group was lower than normal pregnancy model group (P<0.05). Western Blot analysis indicated that the expression of TSLP in chorion and decidua spontaneous abortion model group was lower than normal pregnancy model group (P<0. 05). The expression of TSLP and IL-4 in spontaneous abortion model group was lower than in normal pregnancy model group( P<0.05), but the expression of IFN-y was no significant change than in normal pregnancy model group (P>0.05) ;The expression of CD4 +CD25 +Treg and FoxP3 in spontaneous abortion model group was significant lower than in normal pregnancy model group (P <0. 05). Conclusions:The obvious decrease of TSLP might be an important factor for the disturbance of fatal-maternal immune-tolerance.
