Abstract

Objective: To investigate the effects of anti-oxidant cysteine on in vitro maturation and development potential of mouse oocytes for further optimizing the system of oocytes matured in vitro. Methods: One hundred and one GV oocytes of mouse were cultured in the in vitro culture based medium which consisted of HTF +0. 075 IU/ml HMG+0. 5 IU/ml hCG +1 μg/ml E2 in control group. One hundred and fifteen GV oocytes of mouse were cultured in the medium mentioned above plus 0. 6 mmol/L cysteine in the experimental group. The oocytes in both groups were cultured for 16-18 hours, and then the maturation rates, glutathione levels and mitochondrial membrane potential were observed. Results.- The oocytes maturation rate in the experimental group 80. 0% (92/115) was significantly higher than that in the control group [68. 3% (69/101) ](P<0. 05). The average glutathione levels in the content of oocytes of the experimental group were 0. 51 ±0. 5 μg/L and slightly higher than those in the control group (0. 48i0. 6 μg/L) but there was no statistically significance (P>0. 05). The mitochondrial membrane potential in the experimental group was 0. 39 ±0. 02△Ψm and significantly higher than that in the control group (0. 04±0. 003△Ψm) (P<0. 05). Conclusions; Anti-oxidant cysteine played a significant protective effect on in vitro maturation and development potential of mouse oocytes.
