Establishment and biological characterization of human esophageal squamous cell carcinoma cell line RJEC-2

Abstract
Objective To establish a novel cell line of human esophageal squamous cell carcinoma (ESCC) and to investigate the biological characterization in pursuit of a new model for further studies on molecular mechanisms and therapeutic intervention. Methods Small tissue blocks taken from resected specimens of an ESCC patient were cultured, and squamous cell carcinoma cells were purified. Biological characters of the cell line were investigated, including morphology, cytokeratin expression, growth kinetic features, cell cycle, cytogenetic features and tumorigenicity. Results An ESCC cell line (RJEC-2) was established. It grew continuously in vitro for more than 4 months and 46 passages. This cell line presented features of squamous epithelial cells with growing as a monolayer, cytokeratin positive as indicated by immunohistochemical staining, tonofilaments and desmosomes under electron microscopy. The cells grew with doubling time of 45.5 h, and cell cycle distribution was 56.72% for Q/G,, 33.96% for S, 9.32% for GVM phase when cells grew to 90% confluence. The cytogenentic analysis demonstrated chromosome abnormalities in both structure and number with features of tumor cells. The cell line formed colony with efficiency of 13.93% in culture plates and formed tumors in nude mice. The pathological analysis suggested that the xenograft was similar to the primary tumor and both were moderately-well differentiated squamous cell carcinoma. Conclusions We have successfully established a human ESCC cell line (RJEC-2), a new model for further study on esophageal carcinoma.关键词
