Altered expression of selenium-binding protein 1 during endometrial window of implantation in patients with endometriosis-associated infertility

Abstract
Objective: To explore the association of altered expression of selenium-binding protein 1 ( SBP1) during endometrial window of implantation (WOI) in patients with en-dometriosis- associated infertility and endometrial receptivity. Methods: Proteomic technology, a combination of the two-dimensional fluorescence differential in-gel electrophoresis(2D-DIGE) and matrix-assist laser desorption ionization-time of flight-mass spectrometry ( MALDI-TOF-MS), was adopted to search for proteome alternations of WOI [ at Day-6 after ovulatory day (OD) ,OD+6 day]in patients with endometriosis- associated infertility versus those with tubal infertility. The semi-quantitative validation of SBP1 in the eutopic endometrial tissue during WOI was analyzed by Western blot. Results;By comparing protein profiles,we found a protein, with a isoelectric point of 5. 93 and relative molecular weight of 52. 9 kD, up-regulated 1. 58 times in samples of WOI in patients with endometriosis-associated infertility. Using mass spectrometry, it was identified as SBP1. Protein level of SBP1 was up-regulated during WOI in pa- tients with endometriosis-associated infertility comparing with that in control group by Western blot. Relative intensity of SBP1 were 13.89±1.63 and 11. 34±0. 95(t = -2. 709,P = 0. 035). Conclusions: It seems that the increased expression of SBP1 during WOI in patients with endometriosis-associated infertility plays an important role in the decrease of endometrial receptivity.关键词
