Abstract

OBJECTIVE: To investigate the preventative effects of trimetazidine on gentamicin (GM)-induced nephrotoxicity in rats. METHODS: The rats were divided into solvent control group, trimetazidine low-dose and high-dose groups (5, 10 mg·kg‐1· d‐1), gentamicin group (100 mg·kg‐1·d‐1), trimetazidine (5 mg‐·kg‐1·d‐1)+gentamicin (100 mg·kg‐1·d‐1) group and trimetazidine (10 mg·kg‐1·d‐1)+gentamicin (100 mg·kg‐1·d‐1) with 6 rats in each group. Trimetazidine was administered intragastncally for 2 days; from the third day, gentamicin was given intraperitoneally 30 min after intragastric administration for consecutive 7 days. Other groups were given normal saline accordingly. Rats were sacrificed 24 h after the last administration, and the concentration of gentamicin in renal tissue of rats was determined by fluorescence polarization immunoassay. The apoptotic renal tubular epithelial cells were examined by TUNEL method. Morphologic changes of kidney were determined by HE staining. RESULTS: Compared with gentamicin group, the concentrations of gentamicin in renal tissue of trimetazidine (10 mg·kg‐1·d‐1)+gentamicin group decreased significantly (P<0.05); the numbers of apoptotic renal tubular epithelial cells in trimetazidine (5 mg‐·kg'-d^+gentami-cin group and trimetazidine (10 mg‐·kg‐1·d‐1)+gentamicin group decreased significantly (P<0.05 or P<0.01). The inflammatory cell infiltration and pathology were found in renal tissue of rats in gentamicin group. Renal pathology of renal tissue was improved in trimetazidine (5 mg·kg‐1·d‐1)+gentamicin group and trimetazidine (10 mg·kg‐1·d‐1)+gentaniicin group. CONCLUSION: Trimetazidine can protect the rat against gentamicin-induced nephrotoxicity by decreasing the accumulation of gentamicin in kidney.
