Abstract
OBJECTIVE: To study the protective effect of ligustrazine combined with tanshinone II a on lung injury in rats with ischemia-reperfusion injury. METHODS: SD male rats were divided into 7 groups, I.e. Blank control group (constant volume of normal saline), sham operation group (constant volume of normal saline), model group (constant volume of normal saline), dexa-methasone group (5 mg·kg-1), TMP+tanshinone ⅡA high-dose, medium-dose and low-dose groups (40+12 mg·kg-1, 20+6 mg-kg-1, 10+3 mg·kg-1). Those groups were given relevant medicine intragastrically once a day 3 days before ischemia. MDA content, MPO activity, wet weight/dry weight ratio {WID) and pulmonary permeability index were determined. RESULTS: Compared with blank control group, MDA content of model group increased significantly (P<0.01), MPO activity of lung homogenate was significantly enhanced (P<0.01) , WID, pulmonary ventilation index increased significantly (P<0.01). Compared with model group, plasma content of MDA in TMP+tanshinone Ⅱ a high-dose and middle dose groups was decreased significantly (P<0.01 or P<0.05), lung homogenate MPO activity was significantly decreased (P<0.01 or P<0.05), WID and pulmonary ventilation index decreased significantly(P<0.01 or P<0.05). CONCLUSION: Combined therapy of ligustrazine+tanshinone ⅡA has a protective effect on lung ischemia-reperfusion rats, its mechanism is related to reduction of peroxides metabolites, reduction of neutrophils generation, increase of pulmonary capillary permeability.关键词
