摘要

Abstract

Aim To develop a HPLC method for the determination of plasma concentration of Gambogenic acid (GNA) and study the pharmacokinetics of GNA in rat plasma. Methods A HPLC method was established for the determination of GNA in rat plasma using gambogic acid as the internal standard. The plasma concentration of GNA in rats was determined by HPLC after intravenous administration of GNA at a dose of 1, 2 and 4 mg · kg-1 , and the pharmacokinetic parameters were calculated by BABP 3. 0. Results Calibration curve was linear over the range of 0. 07 ~ 18. 00 mg · L-1 . The lower limit of quantification was 0. 07 mg · L-1 . The extraction recoveries were higher than 70% . The intra- and inter-day precision ( RSD% ) was lower than 10%. This validated method was successfully applied to a pharmacokinetic study after iv administration of GNA in rats at a dose of 1, 2 and 4 mg · kg-1. The T2/1was(43.61 ±0. 61) ,(46. 07 ±2. 11) and (46.73 ± 1.14) min, respectively, and the AUC0-t was (158.52 ±6. 27), (211. 13 ±9.33) and (361.37 ± 14.97) min · mg · L-1, respectively. Conclusion A simple, rapid, accurate and precise HPLC method for the analysis of GNA in rat plasma is successfully developed and validated and successfully applied to a pharmacokinetic study of GNA in rats.
