MTT法在木醋杆菌活力检测中的应用
Application of MTT in the Determinhation of Bacillus xylinus Vitality

Abstract
[Objectives} To determine the vitality of Bacillus xylinus during static culture by MIT assey, bo as to explore a rapid method tó detect the growth and reproduction of Bacillus xylinus. [ Method] With Bacillus xylinus as the study object, the feasibility of MTT melhod in counting the bacteria and measuring ihe bacterial quantity was discussed, and the effects of reaction time, MTT quantity and centrifugal force on the MTT determination results were studied. [ Result] The optical density al 570 nm was positively correlated with the concentration of bac-teria when the viable number of the bacteria was in the range of 3. 90 × 10s - 1. 25 × 10 cfu/ml. The best experimental conditions of MTT methods were 7 000 g centrifugal force, 4 h reacting time, 30 μl addition quantity of MTT, and DMSO as solvent before determination. [Con-clusion] MTT colorimelric melhod can quickly and easily measure the number of viable Bacillus xylinus.关键词
