摘要

Abstract

Objective To investigate the hepatoprotective effect of total triterpenoids of Prunella vulgaris L. ( TTP ) on rats. Methods Sixty rats were randomly divided into six groups as follows: control group, CC14 group, TTP ( 62. 5 , 125 and 250 mg/kg ) groups and bifendate ( 200 mg/kg ) group. Rat acute liver injury model was induced by peritoneal injection of 50% CC14( 2 ml/kg, dissolved in corn oil ). Rats in therapeutic groups were pretreated with TTP or bifendate six days before CC14 was given. The effects of TTP on serum levels of alanine aminotransferase ( ALT ) and aspartate aminotransferase ( AST ), hepatic malondialdehyde ( MDA ) content, glutathione peroxidase ( GSH-Px ) and superoxide dismutase ( SOD ) activities in different groups were investigated. In addition, his-topathologic changes of the liver were detected with hematoxylin-eosin ( HE ). CYP2E1 protein and mRNA expressions in liver tissue were evaluated by Western blotting, immunohistochemical staining and transcriptase-polymerase chain reaction, respectively. Results TTP effectively decreased serum ALT, AST levels, hepatic MDA content, and restored hepatic SOD and GSH-Px activities in acute liver injury rats. Furthermore, TTP reduced CYP2E1 expression at both protein and mRNA levels in a dose-dependent manner. Conclusion TTP had protective and therapeutic effect on rat liver injury, which might be associated with its antioxidant properties and inhibition of CYP2E1 activation.
