Effect of 7, 3′-dimethoxy hesperetin on immune function in rats with adjuvant arthritis

Abstract
Objective To investigate the effect of 7, 3'-dimethoxy hesperetin ( DMHP ) on immune function in rats with adjuvant arthritis ( AA ). Methods Freund's complete adjuvant was used to induce AA in rats. DMHP ( 22, 44, 88mg/kg ) were administered intragastrically on 12th day after AA induction, once a day from day 12 to 21. Splenic lymphocyte proliferation response induced by concanavalin A was examined with MTT assay. IL-2 production of splenic lymphocytes and IL-1 , IL-10, TNF-a productions of peritoneal macrophage ( PMΦ) ) were all determined by enzyme linked immunosorbent assay ( ELISA ). In addition, the levels of TNF-a, IL-1 , IL-10 mRNA were determined by using RT-PCR. Results The suppressed lymphocyte proliferation and IL-2 production of splenic lymphocytes in AA rats were reversed by treatment with DMHP ( 44 , 88 mg/kg ). Meanwhile, DMHP could down-regulate IL-1 , TNF-α productions of PMΦ) and up-regulate IL-10 production of PMΦ at both protein and transcription levels in different degrees. Conclusion One possible mechanism of the therapeutic effect of DMHP on AA rat may be related to adjusting abnormal immune function in AA rats and keeping the balance of cytokine network.关键词
