RT-PCR驱动蛋白Eg5在肝细胞癌中的表达及其意义
The expression and significance of kinesin Eg5 in hepatocellular carcinoma
摘要
Abstract
Objective To detect the expression of kinesin Eg5 in hepatocellular carcinoma ( HCC ), and to assess its correlation with metastasis in HCC. Methods Immunohistochemistry was used to detect expression of Eg5 in cancerous tissues from 53 cases of HCC and 5 cases of normal liver tissue. The correlation between Eg5 expression and clinicopathologic features and metastasis were also analyzed. The expression of kinesin Eg5mRNA in inflammatory pseudotumor liver tissue and without( PVTT)liver cancer tissues and within( PVTT)liver cancer tissues was examined by semi-quantitative reverse transcription-polymerase chain reaction ( RT-PCR ). Results Eg5 was not seen obvious in normal liver tissues, The positive rate of Eg5 expression in HCC tissues were 62. 3%( 33/53 ), and the expression level was significantly correlated with tumor size( x2 = 10. 162, P =0. 002 ), Edmondson grading ( X2= 4. 414 ,P = 0. 038 ), TNM stage( x2 = 13. 061 ,P> = 0. 000 )and whether PVTT( x2 = 4.098 ,P = 0. 041 ). No significant correlation was found with gender, age and whether tumor diolame or not. In comparative analysis, the expression levels of Eg5mRNA in without-PVTT liver cancer tissue and within-PVTT liver cancer tissue were 0. 647 ± 0. 028 and 0. 933 ±0. 017 respectively ( P <0. 05 ), whereas in inflammatory pseudotumor liver tissue the expression of Eg5 mRNA was not found. Conclusion Eg5 is not obviously expressed in normal liver tissues, but shows high expression in HCC tissues. The expression of kinesin Eg5 is associated with metastasis of HCC. Eg5 may be a potential target of HCC chemotherapy.关键词
