熟地、鳖甲对大鼠成骨细胞增殖、碱性磷酸酶蛋白和核心结合因子α1 mRNA表达的影响
Influences of Shudi (Radix Rehmanniae Praeparata) and Biejia ( Carapax Trionycis ) on osteoblast proliferation, and expressions of alkaline phosphatase protein and Cbfα1 mRNA in rats

Abstract
Objective To observe the influences of medicinal serum of Shudi (Radix Rehmanniae Praeparata) and Biejia (Carapax Trionycis) on the proliferation and function of osteoblasts in rats. Methods SD rats were randomly divided into 3 groups including high-dose medicinal group (high-dose group) , low dose medicinal group (low-dose group) and blank group. High-dose group and low-dose group were intragastrically given decoction of Shudi and Biejia (3.250 g/kg and 1.625 g/kg) respectively twice a day and blank group was given equivalent distilled water. The medicinal serum samples of Shudi and Biejia and blank control serum samples were prepared after 3 days. The rat osteoblasts were collected after primary culturing to third generation and identifying with alkaline phosphatase (ALP). All osteoblasts were divided into 3 groups after counting and plating and treated for 72 hours. The proliferation rate of osteoblasts were detected by using MTT test, secretion of ALP was detected by using ELISA and corrected with corresponding OD value, and mRNA expression of core-binding factor al ( Cbfal ) was detected by using RT-PCR in all groups. Results The proliferation rate of osteoblasts, concentration of ALP and expression of Cbfal mRNA were all higher in low-dose group than those in other two groups. Conclusion Shudi and Biejia can protect bone through promoting the proliferation proficiency of osteoblasts关键词
