Abstract
Objective: To observe the influence of azithromycin in the expression on peripheral blood mononuclear cells ( PBMC) direved-dendritic cells(DCs)in asthmatic children, and to investigate the immunomodulating effect of azithromvcin. Methods: Sixteen asthmatic children and 18 healthy children were selected. Peripheral blood mononuclear cells ( PBMC) were isolated from venous blood by density gradient centrifugation under aseptic conditions. Different concentrations of azithromycin (0 mg/L, 0.1 mg/L and 10.0 mg/L) were added into the cultures in asthmatic group. The control group was not interfered with azithromycin. Then PBMC samples were induced with rhGM-CSF. CD86 .CD80 and CD83 were examined by fluorescent activated cell sorter, and the levels of IL.-10 and IL-12 in the supernatant were determined by enzyme-linked immunosobent assay. Results; ( 1 )The expression of CD86 in asthmatic group was significantly higher than that in the control group (t'= 2. 27,P<0. OS ). The expressions of the co-stimulatory molecules CD80 and CD83 between the two groups had no differences (t = 1. 17, 1. 34, P>0. 05 ). ( 2 ) The levels of IL-10 and 1L-12 in asthmatic group were significantly lower than that in the control group (t' = 3.31, 3. 39, P<0.01 ). (3) No significant difference was found in the expressions
of CD80, CD86 and CD83 among different concenlrationa of azithromycin in asthmatic group ( q = 0. 031 ~ 1. 022, P>0. 05). (4) The levels ofIL-10 in concentrations of azithromycin at 0.1 mg/L and 10.0 mg/L were significantly lower than that at 0 mg/L (q = 3. 042 or 4.502, P<0. 01 );while there were no significant differences in the levels of IL-10 between 0. 1 mg/L and 10.0 mg/L groups (q=1.460,P>0.05).The level of IL-12 in the 0.1 mg/L group were significantly lower than that in the 0 rag/L and 10 mg/L groups (q = 3. 148 or 3. 940, P<0. 01 ) ; while there were no significant differences between the 0 mg/L and 10 mg/L groups (q = 0. 792, P>0. 05). (5) IL-10 were positively correlated to IL-12 in asthma group (r = 0. 740, P<0. 01 ), while there were no correlations in heallhy control group (r=0.232, P>0.05). Conclusions:(1) DCs dysfunction existed in asthmatic children. It mainly manifested in the increase of the expression of CD86 and the decrease of the secretion of IL-10 and IL-12. (2 )There were no significant effects on the expressions of CD80, CD86 and CD83 on DCs after being interfered with azithromyein, while IL-10 and IL-12 decreased and had azithromycin-concentration dependence. It was suggested that azithromyein played a negative role in immune regulation to DCs.关键词
