摘要

Abstract

Objective To investigate the potential correlation between pathogenesis of primary Sjogren' s syndrome ( pSS ) and the function of monocyte-derived dendritic cells. Methods Monocytes from 11 pSS patients and 11 healthy controls were isolated and induced to dendritic cells in the presence of IL-4 and GM-CSF. Polyinosinic polycytidylic acid ( polyI: C, 50 μg/mL ) was used to simulate the monocyte - derived dendritic cells. Serial cytokines and TLR3 mRNA were assessed by ELISA and RT - PCR, respectively, at the 6th, 12th and 14th hour after polyI: C treatment. Results Significantly more IFN -α, IFN -β and TNF - α were produced in monocyte - derived dentritic cells from pSS patients when challenged with polyI: C. Although TLR3 mRNA expression was down - regulated by polyI: C stimulation, it was impaired in pSS patients. Conclusion The Hypersensitivity to polyI: C, which may associated with TLR3 abnormality , of the monocyte-derived dendritic cells from pSS patients was identified. The abnormality of monocyte-derived dendritic cells from pSS patients may involved in the pathogenesis of pSS.
