Abstract
Objective: To investigate the effect of 5-aminolevulinic acid (5-ALA)-photodynamic therapy (PDT) on ovarian carcinoma cell lines SKOV3 and AO in vitro. Methods: SKOV3 and AO cells were incubated in a medium containing different concentrations of 5-ALA CO. 1,0. 5,1. 0,2. 5.12. 5 mmol/L) for 4 hours, and they were exposed to different light doses (0.1,0.5,2.5 or 12. 5 J/cm2) of semiconductor laser at 630 nra. The survival rate of SKOV3 and AO cells was analyzed by MTT assay, and compared to that of the simple drug-control group (which was incubated with various concentrations of 5-ALA without exposure to laser), simple laser-control group ( which was illuminated with different doses of laser exposure without drug) and negative control group. The half inhibitory concentration UC50) of PDT on AO and SKOV3 cells was calculated. Results: After 5-ALA-PDT treatment, the survival rates of both ovarian carcinoma cell lines were significantly lowered (P<0. 05), suggesting that 5-ALA-PDT could kill ovarian carcinoma cell lines SKOV3 and AO in vitro, and the killing effects showed concentration-dependant on 5-ALA and energy density-dependant on laser, until the concentration of 5-ALA reached 2. 5 mmol/L or the light doses reached 2. 5 J/cm2. Compared with the negative control group, the changes in cell survival rates of simple drug-control group and simple laser-control group were not significant (P>0. 05). The half inhibitory concentration of PDT on AO and SKOV3 cells was 0. 34 mmol/L and 2. 50 mmol/L respectively, the difference was statistically significant (P<0. 05). Conclusions: 5-ALA-PDT may effectively kill the SKOV3 and AO cells in vitro, and the effect on SKOV3 cells is stronger than on AO cells.关键词
