Abstract

Objective To prepare a molecularly imprinted monolithic column for selectively enriching methotrexate( MTX) in human serum in order to facilitate the RP-HPLC analysis. Methods Melamine was employed as an analogue template to prepare the molecularly imprinted monolithic column by in-situ polymerization. Methacrylic acid was used as a function monomer and ethylene glycol dimethacrylate as a cross linker during the polymerization process. The recognition ability of the imprinted monolithic column towards MTX was evaluated by HPLC analysis. Results The matrix impurities in human serum could be eliminated and MTX was retained on MIP column during consecutive elution with methanol-water (1:9) and acetonitrile-water-acetic acid(96:3.5:0.5). Finally,MTX was obtained through the elution process of methanol for HPLC analysis. Conclusion The prepared MIP monolithic column showed good selective recognition capacity towards MTX under the optimal conditions of mobile phase,which may be applied in online or off-line manner to selectively enrich MTX in human serum, and clear the matrix impurities to provide good MTX fraction for RP-HPLC analysis.
