壮药柿叶槲皮素和山柰素含量测定方法的建立
Establishment of Content Determination of Quercetin and Kaempferol in Zhuang Medicine of Persimmon Leaves
摘要
Abstract
Objective To investigate the method for content determination of quercetin and kaempferol in persimmon leaves. Methods To research the chromatographic conditions of high performance liquid chromatography ( HPLC) , then the orthogonal design was applied to study the samples' preparation conditions. Results Chromatographic condition as follows : Hypersil ODS2 C18(4.6 mm ×250 mm,5 μm) , the mobile phase consisted of methanol -0.4% phosphoric acid(50 '?50) with the flow of 1.0 ml/min,and the detection wavelength was 360 nm. The samples' preparation conditions as follows; The fragmentation degree was 60 mesh, extraction solvent was methanol -25% hydrochloride (4:1) ,solvent inventory amounted to 10 ml,reflux time was 1 hour. Conclusion The HPLC method we established is accurate and reliable , which can be used for determination of quercetin and kaempferol in Zhuang medicine of persimmon leaves.关键词
