摘要

Abstract

To explore the effect of (he invasion function of bladder cancer cell T24 after gene silenced integrin linked kinase(ILK) gene. ILK miRNA(micro RNA) were constructed, then transfected into bladder cancer cell lines T24. The change of ILK gene and protein was delected by RT-PCR (reverse transcriptase-poly-raerase chain reaction) and Western blot. The change of invasion function were measured by heterogeneous adhesion experiment and Transwell. The ILK gene and protein was decreased after ILK miRNA was transfected. The A value of heterogeneity adhesion was 0.21 ±0.05 in miRNA plasmid group. The A value of heterogeneity adhesion was 0.67+0.08 and 0.63±0.04 in negative plasmid group and untransfected group (P< 0.05). The cell number of invasion was (7.2±1.1) cells/HP in miRNA plasmid group, but in negative plasmid group and untransfected group was (15.4±1.6) cells/HP and (13.2±1.2) cells/HP (P<0.05). The results indicated that silencing ILK would decrease the invasion fuction of bladder cancer cell line T24.
