荧光金逆行标记乳鼠视网膜神经节细胞的实验研究
Research on retinal ganglion cells of neonatal rats labeled retrogradely with fluorogold
摘要
Abstract
Objective To establish a method to label the retinal ganglion cells of neonatal rats with fluorogold. Methods One-day neonatal Sprague-Dawley rats were anesthetized by intraperitoneal injection with chloral hydrate, and then 1 μ1 of 4% fluorogold was mi-croinjected bilaterally into the superior colliculi of rats. Four days later, primary mixed retinal cell cultures were established from retinas of five-day neonatal rats. Results Under fluorescence microscope, retinal ganglion cells in mixed cultured retinal cells were successfully labeled with fluorogold, which accounted for 0. 34% of total retinal cells. In addition, retinal ganglion cells survived for three weeks in vitro. Conclusion Retinal ganglion cells of neonatal rats labeled by fluorogold is a feasible approach, and could contribute to the I-dentification of retinal ganglion cells in primary mixed retinal cell cultures.关键词
