Genetic Relationship of 27 Walnut Germplasm Resources Based on ISSR Markers

Abstract
ISSR technique was applied to analyze the genetic relationships of 27 walnut germplasm resources from 4 species. Ten primers with excellent reproducibility and limpid bands were screened out from 120 ISSR primers to amplify the genome DNA of 27 germplasms. 129 bands were amplified out totally, in which 91 were polymorphism bands, accounting for 70. 54% of the total. The value of Nei's genetic similarity index of 27 germplasm varied from 0. 496 1 to 1. 000 0. The results demonstrated a relatively rich genetic diversity of the germplasm and genetic relationship of the samples. Cluster analysis with UPGMA method showed that the 27 germplasm could be divided into 4 groups, which was consistent to the morphological classification. The study will provide references for further development and utilization of walnut germ-plasm.关键词
