宫颈上皮内瘤变及宫颈癌患者外周血Treg/Th17细胞及相关细胞因子的检测与临床意义
The Treg/Th17 imbalance in patients with cervical cancer and cervical intraepithelial neoplasia

Abstract
Objective:To explore the role of Treg/Thl7 in the development of CIN and cervical cancer and observe the mechanism of Treg/Thl7 in tumor immune escape of CIN and cervical cancer. Methods; All samples were divided into three groups;group 1;cervical cancer; group 2;cervical intraepithelial neoplasia( CIN);group 3:normal group. The frequencies of Thl7 cells and Treg cells were detected by flow cytometric analysis. The concentration of IL-23、1L-10、IL-17、IL-6、TGF-β in plasma of CIN and cervical cancer patients were detected by enzyme linked immunosorbent assay ( ELJSA). Results;(1) Compared with healthy volunteers, the frequencies of Treg were significantly elevated in the cervical cancer and CIN patients (P<0.05), and cervical cancer patients have higher frequencies of Treg in the PBMCs than CIN patients( P <0.05). (2) Compared with healthy volunteers, the frequencies of Thl7 were significantly decreased in the cervical cancer and CIN patients (P<0. 05) ,and cervical cancer patients have lower frequencies of Thl7 in the PBMCs than CIN patients(P<0.05). (3)The cytokines levels in plasma detected by ELJSA indicated both TGF-β and IL-10(Treg related)were higher in cervical cancer and CIN patients than in healthy volunteers (P<0.05) ,in addition, cervical cancer patients have higher concentration of TGF-p and 1L-10 in the plasma than CIN patients (P< 0.05). (4)The cytokines levels in plasma detected by ELJSA indicated IL-23,IL-17 and IL-6 (Thl7 related) were lower in cervinal cancer and CIN patients than in heallhy volunteers(P<0. 05) ,in addition,cervical cancer patients have lower concentration of IL.-23,IL-17 and IL-6 in the plasma than CIN patients (P<0. 05). Conclusions: This imbalance of Treg cells to Thl7 cells play an important role in the development of CIN and immune escape of cervical cancer, the mechanism may be relevant with the regulation of cytokines of Treg,Thl7 in the tumor immune microenvironment. Treg/Thl7 balance will provide a new target for the treatment on CIN and cervical cancer.关键词
