疏肝健脾方对NASH大鼠肝组织IKKβmRNA和蛋白表达的影响
Effects of Shu-gan Jian-pi prescriptions on expression of IKKβ at mRNA and protein levels in liver tissues of rats with non-alcoholic steatohepatitis

Abstract
AIM: To invesligale the effects of Shu — gan Jian — pi prescriptions on the expression of inhiblor kB kinase β (IKKβ) at mRNA and prolein levels in the liver tissues of rats with non — alcoholic slealohepalilis (NASH). METHODS: The experimental model of NASH was established by feeding a high — fat diet in rals. The Shu — gan Jian — pi prescriptions of tradilional Chinese medicines were prepared and divided into high dose and low dose of Shu — gan recipes, Jian — pi recipes and composite recipes. After intragslrically trealed wilh the recipes for 16 weeks, the rals were exe- cuted Lo oblain blood samples from the ventral aorta. The serum levels of total cholesterol (TC) , Lriglyceride (TG) , high — density lipoprotein cholesterol (HDL — C) and low — densily lipoprolein choleslerol (LDL — C) were delecled by an au-lomalic biochemical analyzer. The histopathological changes of the liver tissues were observed under microscope with HE slaining. The serum levels of TNF — α, IL — 1 and IL — 6 were determined by ELISA. The mRNA expression of IKKβ in liver Lissues was analyzed by real — Lime fluorescence quanLiLaLive RT — PCR. The proLein level of phospho — IKKβ ( p — IKKβ) in Lhe liver Lissues was examined by WesLern bloLLing. RESULTS: The hisLopaLhological changes of Lhe liver Lissues in Lhe raLs fed wiLh high — faL dieL for 16 weeks indicated Lhe successful esLablishmenL of NASH model. Compared wiLh normal group, Lhe serum TC and LDL — C, liver TC and TG levels,and Lhe serum levels of TNF —α, IL — 1 and IL — 6 in model group were significanLly increased and Lhe expression of IKKβ mRNA and phospho — IKKβ proLein in model group were higher . Compared wiLh model group, Lhe levels of serum TG, and liver TC/TG in Shu — gan recipe high dose group, Jian — pi recipe high dose group and composiLe recipe high and low dose groups were significanLly decreased. The levels of TNF —α, IL — 1 and IL — 6 in Lhe hepatic homogenaLe in composiLe recipe high dose group and Jian — pi recipe high dose group were significanLly decreased. The expression of IKKβ mRNA and p — IKKβ proLein in composiLe recipe high and low dose groups and Jian — pi recipe high dose group were also decreased. CONCLUSION: The increasing levels of inflamma-Lory factors and high expression of liver IKKβ mRNA/p — IKKβ protein play roles in the development of NASH. Reducing the accumulation of inflammatory factors and down — regulation of liver IKKβ may be an important mechanism of Shu — gan Jian — pi prescriptions to prevent and cure NASH.关键词
