LC-MS/MS测定大鼠血浆中乌头碱的浓度
Determination of aconitine concentration in rats plasma by LC-MS/MS

Abstract
AIM: To establish an LC-MS/MS method for determining the concentration of a-conitine in rats plasma. METHODS: Verapamil was used as internal standard, the plasma samples extracted by MTBE were separated on a HyPURITY Cyano column ( 150 mm X 2. 1 mm, 5 μm) with mobile phase contained acetoni-trile and 10 mmol/L ammonium formate (70: 30, V/V) at flow rate of 0.30 mL/min. The electrospray ionization (ESI source) in positive ion multiple reaction monitoring (MRM) were used for scanning analysis, then, aconitine and verapamil ion selective channels were: m/z 646.4→586.4 and 455. 2→164. 9, respectively. RESULTS; The linear range for aconitine was 9. 3 — 2390 pg/mL. The lower limit of quantita-tion was 9. 3 pg/mL, and the relative standard deviation of intra-day and inter-day were less than 15%. CONCLUSION: The established method is suitable for pharmacokinetic study of aconitine.关键词
