类风湿关节炎大鼠滑膜组织中凋亡因子Bcl-xl和Bcl-2及Bax的表达及意义
Expressions and Significance of Apoptosis-Related Factors Bcl-xl,Bcl-2 and Bax in Synovial Tissues of Rats with Rheumatoid Arthritis

Abstract
Objective To explore the changes of the expressions of apoptosis - related factors Bel - xl, Bel - 2 and Bax in the synovial tissues of rats with rheumatoid arthritis ( RA ) in order to further understand the pathogenesis of RA. Methods 16 male Wistar rats were randomly divided into control group and RA model group induced by collagen after one week of adaptability breeding, with each group 8 rats. The rats were killed six weeks after primary immune. Immunihistochemistry, RT - PCR and Western blotting were used to detect the changes of the expressions of apoptosis - related factors Bel - xl, Bel - 2 and Bax in synovial tissues of RA model group and control group. Results The expression levels of Bel - xl and Bel - 2 mRNA in synovial tissues of RA model group were significantly higher than those of the control group ( P <0. 01 ) .A tiny of positive expression of Bax mRNA could be detected in synovial tissues of the control group, but the expression level was still significantly lower than that of the RA model group ( P < 0. 05 ) . Meanwhile, Bel - xl, Bel - 2 and Bax protein all had a small amount of expressions in the synovial tissues of control group, but the expression levels of these three factors were much more significant in RA model group, in which the expression levels of Bel - xl and Bel - 2 were significantly increased ( P < 0. 01 ), while the expression level of Bax were higher than that of the model group, but showed no statistically significant difference ( P > 0. 05 ) . Conclusion In the pathogenesis of RA, there is abnormally high expression of apoptosis inhibitors Bel - xl and Bel - 2. While the increase of expression levels of gene and protein of apoptosis promoter Bax are significantly lower than the increase of apoptosis inhibitor Bel -xl and Bel - 2, resulting in the imbalance of proliferation/apoptosis in synovial tissues and the occurrence and development of RA.关键词
