噻唑橙光化学法病原体灭活对红细胞质量的影响
The quality of RBC treated with thiazole orange and light pathogen inactivation
摘要
Abstract
Objective To evaluate the effects of thiazole orange (TO) and light pathogen inactivation treatment on the properties of RBC. Methods RBC was treated to with TO and light pathogen inactivation. For control(untreated RBC) and treated RBC,pH,hemolysis rate,potassium leakage,ATP and 2,3-DPG concentration were assessed on day 0,7,14,21,28, 35 of storage at ℃. A total of 10 RBC were treated Results Compared with controls,TO phototreated RBC exhibited minimal changes in pH ,2,3-DPC(μmol/gHb) and ATP (μmol/gHb), with pH ,2,3-DPG(μmol/gHb)and ATP(μmol/gHb) decreased form 7.03 ±0. 07,9. 43 ±1.14,4.23 ±0.58 on 0 d to 6. 57 ±0.06,0,2.40 ±0.49on 35 d in treated RBC,respectively; from 6. 95 ±0.09,10. 67 ± 1. 17,5. 18 ±0. 64 onO d to 6.57 ±0.07,0,2. 99 ±0.44 on 35 d in control RBC,respectively. After 35 d of storage,56.1% and 57. 5% of the initial level were retained in treated RBC and control RBC,respectively. Treated RBC exhibited only slightly increased hemolysis rate (0.27% ±0.06%) than control (0. 11% ± 0.04% ) at the end of 35 d storage .which still met the standard requirement of ≤0. 8%. Treated RBC exhibited moderately increased potassium leakage than control,with (40.41 ±2. 94)mmol/L and (28. 33 ±3. 34)mmol/L potassium at 35 d,respectively (P<0. 01) Conclusion Phototreatment with thiazole orange has little affects for the RBC properties during the storage.关键词
