摘要

Abstract

Objective To study the M007-mediated photodynamic inactivation be used to inactive tumor cells in RBC reclaimed in cancer surgery. Methods One experiment group and three control groups were eet: 1) Experiment group (M007-PCT): RBC plus M007 with irradiation;2) Pbotosensitizer group (M007): RBC plus M007 with no light; 3) Irradiation group (L) : RBC with irradiation;4) Control group (C) : with RBC only. The drug dose and light energy density ranging between 4 and 40μmol/L, and from 2. 16 to 6.48 J/cm2, respectively, were varied. RBC hemolvsis rate were e-valuated following these treatments. Results RBC of group M007-PCT and Group M007 hemolysiaed at the light dose of 2.16 J/cm2. M007 dose ranged from 4 to 40 μmol/L, the RBC hemolysis rates of group M007-PCT and group M007 increased from (0.108 ±0.003)%, (0.092 ±0.001)% to( 1.032±0.074)%,(l. 022 ±0.053)% respectively, dramatically lager than that of group L and C (P < 0.05 ). The light dose ranged from 2.16 J/cm2 to 6.48 J/cm2, hemolysis rate of group M007-PCT (28, 32, 36, 40ujnol/L) increased from (0.920 ± 0. 042)% 、(0.945 ± 0. 054)% 、(1.035 ± 0.052)%,(1.032 ±0.074)% to (1.489 ±0.315)% ,(1.506 ±0.110)% 、( 1.484 ±0. 308)% ,(1.502 ±0.264)%. Conclusion The level of M007-PCT response and the RBC hemolysis rate shows a strong dependence on the drug dose and light energy density in the applied conditions. RBC hemolysis slightly occurred in samples plus M007 without red light, showing dependence on the drug dose too (r = 0.956 、r = 0.946). The dose of experiment group is less than 1.7% hemoly-sis rate.
