Abstract

One hybridoma cell line secreting monoclonal antibody(McAb) 2B10 was produced by fusing mouse myeloma cells(SP2/0) with spleen cells from BALB/c mice immunized with the purified SM-E2-BC protein of classical swine fever virus(CSFV) Shimen strain. The antibody titers were 1 : 600--1 : 1 000. Western-blot and ELISA analysis showed that the McAb 2B10 could react with denatured SM-E2 protein, indicating that the McAb 2B10 could recognize a linear epitope. It did not react with E2 of the C strain or Group 2 strains. The McAb 2B10 could not react with mutant proteins with single alanine substitution at amino acid 707,709,711,713 and 714, further confirming that its specific recognition of a specific epitope 707^I-X-P-X-G-X-G-G^714 on E2 glycoprotein of Shimen strain. This monoclonal antibody may be helpful for the analysis of the antigenic diversity and pathogenesis studies of CSFV.
