Abstract

Aim To investigate the correlation between the anti-proliferation effect of berberine on HCT116 cells and Wnt/β-catenin signaling transduction. Methods Crystal violet staining was employed to detect the anti-proliferation effect of berberine on HCT116 cells. The apoptosis induced by berberine was detected by fluorescence activated cell sorting and Western blot. The p-catenin expression was measured by Western blot and reverse transcription and polymerase chain reaction ( RT-PCT ). With exogenous expression of β-catenin,the role of p-catenin in berberine induced proliferation inhibitory effect on HCT116 cells was explored. Results Berberine could inhibit the prolifera- tion and induce apoptosis in HCT116 cells. The expression of β-catenin protein in whole cell, cytoplasm and nucleus was inhibited when treated HCT116 cells with different concentrations of berberine. So was the mRNA expression level of β-catenin. Exogenous expression of β-catenin could attenuate the anti-proliferation effect of berberine on HCT116 cells. Conclusions Berberine can inhibit the proliferation of HCT116 cells and Wnt/β-catenin signaling transduction. This effect may be mediated at least by down regulating the mRNA expression of β-catenin.
