摘要

Abstract

[ Objective] To isolate and identify a strain of cellulase-producing fungus,so as to provide experimental basis for improving its species diversity. [ Method] The cellulase-producing fungus was firstly screened by Congo red method and then rescreened by flask fermentation from haystack, the fermentation culture medium was optimized by single factor test, and the morphology and 18S rRNA of this fungus were identified. [Result] A strain of cellulase-producing fungus RSC1 was isolated,as indicated by the optimization results of fermentation medium,the activity of produced cellulase was the highest on the medium containing 1.3% CMC-Na,0. 1% yeast extract and 0.1% MgSO4. As shown by the morphology and molecular biology of this fungus,the similarity between RSC1 and Mucor amphibiorum was as high as 97% ,thus the fungus was named as Mucor amphibiorum RSC1. [Conclusion] The study identified the isolated fungus to be Mucor amphibiorum RSC1,which laid foundation for improving the species diversity of cellulase-producing fungus.
