应用生物学软件分析四季橘染色体核型和带型
Analyses of Citrus madurensis chromosomal karyotype and banding style by Biological Image Software
摘要
Abstract
[Objective]This study is to improve the resolution and accuracy of chromosome analysis. [Method]The mitotic metaphase chromosomes of C. madurensis after 4, 6-Diamidino-2-Phenylindole (DAPI) staining were analyzed with quantitative karyotype by MieroMeasure software and Image-Pro Plus software application. [Result]The results showed that: the karyotype formula of C. madurensis was 2n=18= 16 m (2SAT)+2 sin, which belonged to primitive Stebbins 1B type; On the other hand, by using DAPI staining, C. madurensis exhibited variation of the fluorescence intensity in prometaphase chromosomes (from the short arm to the long arm), and based on this, the quantitative DAPI-banding ideogram was constructed. [Conclusion]Chromosome analysis by biological image software showed more detailed karyotype data, which is therefore considered to be a new method for citrus cvtogenetic research.关键词
