嗅鞘细胞体外CFSE染色的标记方法研究

Abstract
Objective: To establish a method of CFSE labeling olfactory ensheathing cells (OECs) at appropriate final concentration in vitro. Methods: OECs were labeled by 2, 5, or 10 μM CFSE in vitro. The positive labeling rate, cell viability, growth and morphological charac teristics and proliferation of OECs were detected by flow cytometry, trypan blue staining and fluorescence microscopy respectively. Results: More than 99% of the OECs were CFSE-labeled at the three concentrations (2, 5, 10 μM) investigated. The OECs labeled with 2 μM CFSE had the highest viability and the maximum proliferation rate. However, the fluorescence intensity was significantly decreased with time. Conclusion: CFSE can be used to label OECs with optimized concentration of 2 μM.关键词
