摘要

Abstract

[Objective]The present experiment was conducted to study the genetic diversity and relationship amongst different water chestnut (Eleocharis tuberosa Schulul) cultivars to provide reference for breeding water chestnut. [Method] Random amplified polymorphism DNA (RAPD) molecular markers were used to assess the genetic diversity amongst 24 water chestnut (Eleocharis tuberosa Schulut) cultivars using RAPD primers and cluster analysis was performed. [Result] Out of 100 primers, 15 primers with clear bands and polymorphism were selected and 83 reproducible DNA bands were amplified, of which 61 bands (73.5%) were polymorphic. The coefficient of genetic similarity in different cultivars ranged from 0.0976 to 0.6757 with an average of 0.3048. UPGMA cluster analysis results showed that all the cultivars were clustered into five groups, and the fourth group could be divided into two subgroups. The wild species belonged to the fifth group alone. [Conclusion ]Narrow genetic diversity and close genetic relationship presented amongst 24 water chestnut (Elmcharis tuherosa Schulut) cultivars, certain genetic differences still existed among those cultivars come from different.
