Abstract

Objective: To explore the establishment of internal control of blood coagulation analyzer by using self-made mixed plasma. Methods: The fresh mixed blood plasma was collected from the patients with normal coagulation, divided into EP centrifuge tubes and then restored in low temperature refrigerator ( — 20 'O for conservation. Two tubes were removed from the refrigerator and placed at room temperature per day. After thawing unaffectedly, one was used as normal blood plasma, another was diluted with normal saline by three folds as abnormal blood plasma. PT, APTT, Fib were assayed simultaneously in the normal and abnormal self-made blood plasma, matching quality control substance and clinical specimens. Results: In three months, there was no statistically significance difference between the cryopreserved self-made mixed blood plasma and the raw self-made mixed blood plasma on the results of PT, APTT and Fib ( P >0. 05). In the third month, PT, APTT and Fib analysis showed the results of the cryopreserved self-made mixed plasma were consistent with those of the matching quality control substance in L-J quality control graph, they had good correlation ( r>0. 95). Conclusion: The cryopreserved self-made mixed plasma can be used as internal control on blood coagulation analyzer. Two tubes were removed per day, one can be used as normal control and the other diluted with normal saline by three folds can be used as abnormal control.
