Construction of RNAi Vectors of Flax Male Sterility Related MS2-F Gene
摘要
Abstract
Flax male sterility related MS2-F gene plays an important role in the process of flax pollen development. In order to conform the function of this gene, two RNAi vectors of the gene were constructed. Two different fragments were selected and inserted into pHANNIBAL flanking PDK intron in sense orientation and antisense orientation , thus forming hpRNA cassettes. Then the Not I fragment from pHANNIBAL containing the hpRNA cassettes were subcloned into the binary vector pART27. Finally the vectors were introduced into Agrobacterium LBA4404. These successfully constructed RNAi vectors and subsequent genetic transformation will provide useful clues for revealing the function of MS2-F gene during pollen development in flax.关键词
