HPLC对肺结核患者利福平血药浓度的监测
Monitoring of plasma rifampicin concentrations in patients with pulmonary tuberculosis by HPLC

Abstract
Objective To monitor plasma rifampicin concentrations in patients with pulmonary tuberculosis by HPLC. Methods The assay was conducted on C18 GL sciences column (5 μm, 4.6 × 250 mm)with the mobile phase of the mixture of CH3OH-CH3CN-0.04mol/L KH2PO3(30:30:40)at a rate of 1.2 ml/min,the UV detection wavelength was 340 nm. The column temperature was 20 ℃. Results A good linearity was obtained from 0. 30 - 24 μg/ml with a correlation coefficient of 0. 999 5. The average recovery of the assay was 98. 87%. Among 140 patients administrated with 450 mg rifampicin orally or intravenously, the plasma drug concentration ranged 0. 50 - 12. 75 μg/ml in 126 cases, ranged 0. 30 - 0.5 μg/ml in 12 cases and was less than 0.3 μg/ml in 2 cases. Conclusion The method applied in this study is accurate and sensitive, which can be used to monitor plasma rifampicin concentration for rational drug administration in clinical practice.关键词
