Abstract
Objective To investigate the effects of erythropoietin( EPO) on the score of neurological deficits, brain edema and expression of matrix metalloproteinases-9 ( MMP-9) in rats after cerebral ischemia-reperfusion. Methods A total of 150 rats were randomly divided into sham operation group, ischemia group and EPO group. The model of focal brain ischemia-reperfusion was made by occluding the middle cerebral artery( MCA) for 2 h. The rats in ischemia group were intraperitoneally injected 2 ml saline 10 min before reperfusion, and rhEPO (3 000 U/kg) +2 ml saline in EPO group. The score of neurological deficits was determined at 6 h, 12 h,48 h, 72 h ,7 d and 10 d after reperfusion. The water content in brain tissues was measured with Elliot formula and the expression of MMP-9 was examined with immunohistochemical stain. Results hi ischemia group,the score of neurological deficits increased at 6 h,peaked at 48 h.and began to decrease at 72 h;the water content increased at 6 h.and peaked at 24 -48 h;the positive expression of MMP-9 was observed at 6 h after brain ischemia-reperfusion,increased at 12 h,peaked at 48 h.and gradually decreased at 72 h,7 d and 10 d. Compared with ischemia group,the score of neurological deficits,the water content in brain tissues and MMP-9 positive cells decreased significantly in EPO group(P<0.05). Conclusion The results suggest that EPO may protect against cerebral iscbemia-reperfusion injury,which is related to the decrease of expression of MMP-9.关键词
