杆状病毒在气升式反应器中感染策略优化
Optimization of infection strategy for baculovirus in airlift reactor
摘要
Abstract
To get the best conditions for the baculovirus infection of suspension insect cells, an orthogonal design was used to select the optimum conditions, in which the effects of multiplicity of infection (MOI) , time of infection (TOI) .initial cell density(ICD) and dissolved oxygen (DO)on the budded virus (BV)titer and occluded virus (OV) number were evaluated in the HzAMl insect cell-recombinant baculovirus system at a low MOI in airlift reactor. The results show that the best conditions for the infection are MOI of 0.1 ,initial cell density of 2 × 105 cell · mL-1 ,infection at the late exponential phase and DO of 10% .and a maximum BV concentration of 7.943 · 106 TCIDs, · mL-1 is obtained,a maximum OV concentration of 1.68 × 106 OVs · mL-1 is obtained.关键词
