摘要
Abstract
Objective:To study the effect of interfering the expression of ERCC1 by ad-enovirus with hTERT on reversing cisplatin resistance in ovarian carcinoma cell lines. Methods : Recombinant adenovirus were divided into 1MOI, 10MOI, 50MOI, 80MOI to infect SK-OV3 ,SKOV3/DDP and ECV304,while the same dose of empty carrier adenovirus and uninfcct-od cells as control group. The expression of ERCC1 mRNA was detected by RT-PCR and the expression of ERCC1 protein was detected by Western blot in recombinant adenovirus- transfec-tion cells and the control groups. The cisplalin chemotherapeutic sensitivity was detected by MTT assay. Results: (l)The expressions of ERCC1 mRNA and ERCC1 protein in SKOV3 and SKOV3/DDP infected with various doses of Ad-hTERT-ERCCl shRNA were declined compared with uninfected groups (P<0. 01 ). The expressions of ERCC1 mRNA and ERCC1 protein in ECV304 cell and cells transfecled with empty vector did not changed compared with the uninfected groups (P>0.05). (2)Cisplatin sensitivity in SKOV3/DDP was efficiently enhanced after infected with recombinant adenovirus(P<0. 01) ,and was dose-dependent. Conclusion; The adenovirus with hTERT promoter can reverse the resistance of cisplatin by interfering the expression of ERCC1 in SKOV3/DDP.关键词
