Abstract
Objective: To analyze the influence of epidermal growth factor receptor (EGFR) inhibitor AG1478 on the proliferation and epithelial-mesenchymal transition ( EMT) in endometrial carcinoma cells. Methods;The expression of epithelial marker proteins such as;E-cadherin, α-catenin, mesenchymal marker proteins such as;N-cadherin, vimentin and MMP-9, MMP-2 were examined in endometrial carcinoma cells through Western blot. The proliferation inhibitor effect of different concentration AG1478 on endometrial carcinoma cells was detected by MTT. Western blot was applied to investigate the influence of AG1478 on expressions of E-cadherin,α-tenin, N-cadherin, vimentin and MMP-9, MMP-2 in endometrial carcinoma cells. The influence of AG1478 on migration of endometrial carcinoma cells was examined by tran-swell migration assay. Results: Over expressed EGFR resulted in the expression of a decrease in epithelial marker proteins and an increase in mesenchymal marker proteins,MMP-9,MMP-2 in endometrial carcinoma cells( P<0. 05 ) . AG1478 could suppress the proliferation of different endometrial carcinoma cells and cells transfected with EGFR were more sensitive(P<0. 05 ). Increase of expression of epithelial marker proteins and decrease of mesenchymal marker proteins, MMP-9,MMP-2 were observed in endomelrial carcinoma cells after AG1478 treated. This effect was more obvious in cells transfected with EGFR(P<0. 05). The migration of endometrial carcinoma cells was supressed by AG1478 and Ishikawa cells transfected with EGFR were demon- straled more sensitive to AG 1478 (P<0. 05). Conclusion: ECFR inhibitor AG1478 can effectively inhibit the proliferation, epilhehal-mesenchymal transition of endometrial carcinoma cells.关键词
