Abstract

Objective:To exame the expression of CXCR7 chemokine receptor and its ligands in endometrial carcinoma tissues and cell lines, and explore whether estrogen can regulate the expression of CXCR7, Methods: RT-PCR was firstly used to analyze the transcription levels of twenty chemokine receptors in Ishikawa(ERa positive) and AN3CA(ERa negative) cell lines, and the chemokine receptor which was expressed highly was selected . The expression of this chemokine receptor and its ligands in endometrial carcinoma tissues and cell lines were examed by immunocytochemical staining, and the effect of estrogen on the chemokine receptor protein levels was explored by Western blot. Results: (1) GXCR7 mRNA was highly expressed in Ishikawa and AN3CA cell lines. Immunocytochemical staining suggested that CXCR7 and its ligand SDF-1 were expressed in endometrial carcinoma tissues and Ishikawa, AN3 CA cell lines, while no expression of the another ligand I-TAC;(2)In Ishikawa,estrogen could up-regulate the protein level of CXCR7 with the changes of concentration,otherwise in AN3CA,the expression of CXCR7 was not notablely changed by the stimulation of estrogen. Conclusions: CXCR7/SDF-1 is highly expressed in endometrial carcinoma tissues and cell lines. Estrogen can up-regulate the expression of CXCR7. CXCR7/SDF-1 seems to act as an important regulator in the endometrial carcinoma.
