摘要

Abstract

Objective: To investigate the effect of isoflurane on tau truncation in hippocampus of aged rats and the possible mechanism. Methods:Sixteen aged Sprague Dawley(SD) rats (age >18 months) were randomly allocated to either anesthesia group (n = 8) or control group (n = 8). The rats in the anesthesia group underwent isoflurane anesthesia (1.5%- 2%) for 6 h. The rats in the control group inhaled vehicle gas for 6 h. Immunofluorescent multiple labeling was employed to study the immu-noreactivity of Tau-C3,cleaved caspase-3,as well as the colocalization of them. Immunoblotting was used to semi-quantify Tau-C3 and cleaved caspase-3. Results: The density of Tau-C3 positive cells were (5. 6 ±2. 4)/high power fields(HPF) in the anesthesia group and (2. 5 ±1. 2)/HPF in the control group. The relative optical density of Tau-C3 in the anesthesia group was significantly higher than that in the control group. Immunofluorescent multiple labeling did not show colocalization of Tau-C3 and cleaved caspase-3 in 95% of Tau-C3 positive cells. The relative optical density of cleaved caspase-3 in both groups was low and the inter-group difference was not statistically significant. Conclusions: Isoflurane anesthesia can enhance the tau truncation at D421 in the hippocampus of aged rats,and the process is not correlated with caspase-3.
