荧光双染和三维重建技术对蠕形螨的形态观察和功能探讨
Morphological observation on Demodex folliculorum by fluorescent double staining and 3D-reconstruction and discussion on their functions

Abstract
The aim of the present study is to reconstruct the three-dimensional (3D) image of the double stained Demodex folliculorum by using confocal laser scanning microscope (Cl-SM), and to study the function of its primary structures thereby. The specimens were initially stained with fluorescein isothiocyanate (FITC) and then double stained with propidium iodide (PI). Placing the double stained specimens under the CLSM. a 3D image was reconstructed by Imaris 5. 0. by which observation and evaluation of the morphological characteristics of the Demoded folliculorum were conducted. Results showed that the specimens were well combined with FITC and PI. A clear image of the mites' external appearance and the anterior feeding canal were exposed by the high definition 3D pictures. Ifs indicated that the combination of fluorescent double stain and 3D-reconstruction technique is fundamental on the further study of the mite's external morphology as well as its internal structures and functions.关键词
