Abstract

OBJECTIVE: To establish a method for the determination of azithromycin concentration in human plasma. METHODS: The plasma samples were protein-precipitated, and the samples were determined by UPLC-MS. The determination was performed on Waters Acquity UPLC BEH C18 column with mobile phase consisted of acetonitrile-10 mmol·mL‐1 ammonium formate (66:34) at the flow rate of 0.2 mL·min‐1. The protonated ions of analytes were detected in positive lonization multiple reaction monitoring mode (MRM). The mass transition pairs of m/z 748.36→591.10 and 837.34→158.06 were used to detect azithromycin and roxithromycin. RESULTS: The linear range of azithromycin was 2~1000 ng·mL‐1(r=0.999 5), and the low limit of quantita-tion was 2 ng· mL‐1. The average method recovery rates were 102.6% ~108.0% and average extraction recovery rates were 93.2%~102.2%. The intra-day RSD and inter-day RSD were all less than 8%. CONCLUSION: The method is simple, quick and reliable with high specificity and sensitivity, and it is suitable for the pharmacokinetic study of azithromycin.
